The Pole lost 6-4, 5-7, 2-6 to Daria Kasatkina of Russia in the tournament’s round of 16 on Wednesday.
Radwańska, who is currently world No. 11 as ranked by the Women’s Tennis Association, faces a substantial drop in the rankings as a result of the defeat, experts said after the match.
Earlier in the tournament, Radwańska beat Germany's Carina Witthoeft 7-5, 6-3 in the first round and overpowered China’s Zhang Shuai 7-5, 7-5 in the second round.
Radwańska defeated Britain’s Johanna Konta in last year's final in the Chinese capital, and she also won the tournament in 2011.
Poland’s Magda Linette crashed out of the China Open after she was beaten on Monday in the first round by Russia’s Elena Vesnina.
Linette lost the first set 0-6, and pulled out of the second, complaining of an injury.
